SHOX2 Antibody (JK-6E) is a mouse monoclonal IgG2a kappa light chain antibody that detects SHOX2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-SHOX2 antibody (JK-6E) is available as a non-conjugated format. SHOX2, also known as SHOT, is a crucial transcription factor belonging to the homeodomain protein family, which plays a significant role in regulating gene expression during development and differentiation. SHOX2 is characterized by a conserved 60-amino-acid homeodomain that allows binding to specific DNA sequences in the regulatory regions of target genes. SHOX2 exists in two isoforms, SHOX2A and SHOX2B, resulting from alternative splicing, which differ in their N-terminal residues and an alternatively spliced C-terminal exon. Both isoforms contain a C-terminal OAR domain, a motif particularly important for craniofacial development. Expression of the mouse ortholog, Og12X, has been identified in various tissues, including the aorta, female genitalia, and several regions of the developing brain, indicating involvement in critical developmental processes. Notably, aberrations in SHOX2 expression have been linked to Cornelia de Lange syndrome, a multisystem disorder characterized by developmental delays and distinct physical features, underscoring SHOX2′s importance in normal development and potential implications in genetic disorders.

We have detected Shox2 expression via immunostaining on shox2 knockout mice using this antibody. Therefore, its specificity is not KO validated. We want to know the exact immunogen for producing this antibody ao that we can push our research forward.

For Western Blot, is it recommended to use denatured or non-denatured conditions with SHOX2 (JK-6E): sc-81955 antibody?
