SGLT-2 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects SGLT-2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). SGLT-2 (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SGLT-2, a member of the sodium-glucose cotransporter family, plays a crucial role in glucose reabsorption in the kidneys, specifically in the proximal convoluted tubule, where SGLT-2 facilitates the transport of glucose from the renal tubular fluid back into the bloodstream. This process is vital for maintaining glucose homeostasis, as SGLT-2 prevents excessive loss of glucose in urine, which can lead to conditions such as glucosuria. SGLT-2 operates by utilizing the sodium gradient established by the Na+/K+ ATPase pump, allowing for the co-transport of sodium ions and glucose molecules. Dysregulation or mutations in SGLT-2 can lead to familial renal glucosuria, a condition characterized by the presence of glucose in urine despite normal blood glucose levels, highlighting the importance of SGLT-2 in renal function and metabolic health.

Technical Support says the antibody is raised against amino acids 226-268 mapping within an internal region of SGLT-2 in Q&A. However, the data sheat says SGLT-2(D-6) is raised against aa 220-270 mapping within an extracellular region. Which is true?

Thanks for your question. The exact peptide sequence used to raise this antibody is proprietary. However, we can give a 50 amino acid range. Anti-SGLT-2 Antibody (D-6): sc-393350 is raised against amino acids 226-268 mapping within an extracellular domain of SGLT-2 of human origin.

Hi Does SGLT2 antibody detect rabbit SGLT2?

Thank you for your question. We have not tested this mouse monoclonal antibody, SGLT-2 (D-6): sc-393350, for reactivity in rabbit samples. The antibody is raised against amino acids 226-268 mapping within an internal region of SGLT-2 of human origin. Based on sequence analysis, the immunogen sequence does share 91% sequence identity with the published protein sequence for rabbit SGLT2 (see link below as a reference for that sequence). This is a very high degree of sequence identity, and there is a good chance that this antibody would detect rabbit SGLT-2. http://www.uniprot.org/uniprot/P26430

I was wondering if this antibody reacts with mouse SGLT-2. They share 86% identity in the amino acid sequence used as a antigen. If not, could you recommend one for IHC/WB?

Based on the sequence identity with the antigen, it is highly likely that this antibody will also detect the mouse protein. We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. sc-393350: SGLT-2 (D-6) is a mouse monoclonal antibody that is specific for SGLT-2. It is not expected to cross-react with SGLT-1.
