S-100 β chain Antibody (C-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects S-100 β chain protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). S-100 β chain (C-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. S-100 β chain protein plays a crucial role in regulating cellular processes, including cell cycle progression and differentiation, which are vital for maintaining normal cellular function and tissue homeostasis. S-100 proteins are implicated in activating calcium-induced calcium release, inhibiting microtubule assembly, and modulating protein kinase C-mediated phosphorylation, highlighting their importance in signal transduction pathways. S-100 family consists of two subunits, S-100 α and S-100 β, which share approximately 60% sequence identity and can form various dimeric combinations, including two α chains, two β chains, or one of each. S-100 β chain is predominantly localized in cytoplasm and nuclei of astrocytes, Schwann cells, and certain tumors, such as ependymomas and astrogliomas, and is also found in benign nevi and malignant melanocytic tumors, as well as in Langerhans cells in skin. S-100 proteins, along with calbindin and parvalbumin, are particularly significant in neural tissues, while S-100 α and β are also present in various other tissues, including intestine and kidney, underscoring their diverse biological roles.

Hello, I want to use S100B to label human cortical astrocytes in culture. Would S-100 β chain Antibody (C-3) work for ICC/IF my culture with astrocytes ? Also do you recommend other markers other than GFAP for astrocytes ?

Thank you for your question. This monoclonal antibody, S-100 β chain (C-3) sc-393919, is human reactive and should be fine for IF and IHC use on human samples. We have not tested specifically on astrocytes, so I do not have any information specifically regarding that sample. We do not have information regarding optimal astrocyte markers, so I would refer you to the primary literature for that information.

Rated 1 out of 5 by from Disappinting signalApplication: Immunofluorescence in (4% PFA fixed ) paraffin embedded sample of rat colon using heat mediated antigen retrieval with TRIS/EDTA buffer (pH:9, 20min, 92°C) or heat mediated antigen retrieval with citrate buffer (10min, 100°C) and a dilution of 1:50. Conclusion: the antibody shows immunoreactivity using antigen retrieval with TRIS/EDTA buffer only. In this preparation it labels the glial cells exclusively inside the ganglia (myenteric and submucous). Likely, not all the ganglia were labelled.
