    Ribosomal Protein L11 Antibody (2A1) is a mouse monoclonal IgG2b kappa light chain antibody that detects Ribosomal Protein L11 (RPL11) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-RPL11 antibody (2A1) is available as a non-conjugated format. RPL11 plays a crucial role in ribosome assembly and function, essential for protein synthesis in all living cells. RPL11 (2A1) monoclonal antibody localizes to the nucleolus, where RPL11 participates in ribosomal RNA processing and maturation, vital for ribosome formation.     Why is your antibody recognising a 40 kDa protein - RPL11 is about 20 kDa. It should also be predominantly cytoplasmic - as that is where the ribosomes are. My lab works on ribosome biogenesis and I think this antibody should be removed from your stock

    Thank you very much for the question! The full-length Ribosomal Protein L11 of human origin has 177 amino acids. The calculated molecular weight of this protein to be detected by standard WB techniques is expected to be around 20kDa. This is also the weight we refer to on the corresponding product datasheet you can download by clicking the following link: https://datasheets.scbt.com/sc-293224.pdf The WB image we show on this product page is showing a Ribosomal Protein L11 - GST fusion protein with the corresponding higher molecular weight. This is the correct mw size we expected in this control experiment but of course not the size to be detected on endogenously expressed Ribosomal Protein L11. In the IF staining of methanol-fixed HeLa cells our labs have performed this antibody was showing nuclear localization. According to the protein literature we found this protein should be expressed predominantly in the cytoplasma but we also found supporting information about IF localization in the nucleus. Our labs will look further into this and post additional data here on the product page upon availability.

    Why is the RPL11 signal in the nucleus when RPL11 is predominantly a cytoplasmic protein? This suggests to me that the antibody is non-functional

    Can sc-293224: Ribosomal Protein L11 (2A1) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Thank you for your question. Yes, sc-293224: Ribosomal Protein L11 (2A1) is recommended for use in IHC with paraffin-embedded sections.
