Rap1GAP Antibody (D-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects Rap1GAP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Rap1GAP antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rap1 GTPase-activating protein (Rap1GAP) plays a crucial role in cellular signaling by specifically stimulating the GTP hydrolytic activity of the monomeric G protein Rap1, which is essential for regulating various cellular processes, including cell adhesion, proliferation, and differentiation. Rap1GAP is primarily located in the cytoplasm and can translocate to the membrane upon stimulation, which is vital for modulating signaling pathways. Notably, Rap1GAP interacts with Gα z, a member of the Gi family of trimeric G proteins, which inhibits the ability of regulators of G protein signaling to stimulate GTP hydrolysis of the α subunit, thereby influencing downstream signaling events. Rap1GAP expression in the brain, kidney, and pancreas suggests involvement in diverse physiological processes, and the discovery of a novel isoform, Rap1GAPII, which binds specifically to Gα z, highlights the complexity of regulatory mechanisms. Stimulation of the Gi-coupled M2 muscarinic receptor leads to the translocation of Rap1GAPII from the cytosol to the membrane, resulting in decreased levels of GTP-bound Rap1 and subsequent activation of the ERK/MAPK signaling pathway, underscoring Rap1GAP′s importance in integrating G protein signaling and cellular responses.
