Rad9 Antibody (B-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects Rad9 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Rad9 (B-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rad9 protein plays a crucial role in the DNA damage response, particularly in regulating cell cycle checkpoints. Rad9 is primarily located in the nucleus, where Rad9 is involved in forming the Rad9-Rad1-Hus1 complex, which is essential for activating checkpoint kinase Chk1. This complex maintains genomic stability by halting the cell cycle in response to DNA damage, allowing repair mechanisms to occur before proceeding to mitosis. Rad9′s interaction with Chk1 and phosphorylation status are critical for proper G2 checkpoint functioning, preventing damaged DNA replication and reducing tumorigenesis risk. Rad9′s association with anti-apoptotic Bcl-2 family proteins suggests a role in regulating apoptosis following DNA damage, highlighting Rad9′s importance in both cell cycle control and cellular stress responses.

For Western Blot, is it recommended to use denatured or non-denatured conditions with Rad9 (B-8): sc-74464 monoclonal antibody?

Rated 5 out of 5 by from Very strong signal!We have tried this antibody on U2OS cells and we detect a strong signal between 63 and 75 kDa with 1:1000 dilution on western blot after 1 minute exposure on film.

Rated 5 out of 5 by from Clean band at expected heightI have tested the antibody on lysates of human primary breast and lung cells. Using the Turboblot/Odyssey system, I obtained a clean, strong band around ~75 kDa, similar to the picture on the website. I incubated the blots with anti-Rad9 antibody at a 1:100 concentration in 5% BSA O/N.
