SEE ALSO...

Rad52 Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects Rad52 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-Rad52 antibody (F-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rad52 protein plays a crucial role in DNA double-strand break repair, a vital process for maintaining genomic stability and preventing cell death, mutations, or neoplastic transformation. Specifically, Rad52 forms a heptameric ring structure that binds to single-stranded DNA ends, facilitating the annealing of complementary strands, which is essential for effective DNA repair. This structural feature allows Rad52 to efficiently mediate the repair process, particularly in conjunction with other proteins in the Rad52 family, such as Rad51, which interacts with BRCA1 and BRCA2 to influence cellular responses to DNA damage. The interplay between these proteins is critical, as inactivation of BRCA2 can lead to genomic instability and contribute to tumorigenesis. Rad52 monoclonal antibody (F-7) is an invaluable tool for researchers studying DNA repair mechanisms and their implications in cancer biology.

What dilution do you use for IF?

Asked by: platon
Thank you for your question. We used this mouse monoclonal, Rad52 Antibody (F-7): sc-365341, at a dilution of 1:50 for immunofluorescent staining. The recommended range is 1:50-1:500, and optimal conditions should be determined by titrating the antibody in your application. A link to our recommended protocol for IF is provided below. https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

Can you recommend a positive control to use with Rad52 (F-7): sc-365341 monoclonal antibody for Western blot?

Asked by: jenniferc
We recommend HCT 116 (sc-364175), SK-MEL-28 (sc-2236), HeLa (sc-2200), HEK 293 (sc-45136) or Raji (sc-364236) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of Rad52 in these cell lines.

What secondary is best to use with Rad52 (F-7): sc-365341 monoclonal antibody?

Asked by: jenniferc
Thank you for your question. While most anti-mouse IgG secondary antibodies are appropriate for use with Rad52 (F-7): sc-365341, we recommend using one our exclusive Mouse IgG Binding Proteins as a secondary detection reagent. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety

Rated 4 out of 5 by from Good for western blotThis antibody is good for WB. After long term storage, it can get weeker. Longer exposure is recommended.
Date published: 2019-03-03
Rated 2 out of 5 by from Not appropriate for IFThe antibody shows a pan-nuclear staining, but this is regardless of DNA damage treatment. I could not see any damage-specific foci formation.
Date published: 2018-03-16
Rated 4 out of 5 by from Worked well for WBThis Ab worked great for WB. For IF, I did get signal from my staining but did not get nice foci unlike picture shown in website.
