Rad51D Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody designed to recognize the C-terminal epitope encompassing amino acids 233-328 of the human Rad51D protein, ensuring precise detection across multiple applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) in mouse, rat, and human samples. Rad51D, also called TRAD, R51H3, Rad51l3, and Trad-d5, is a critical member of the Rad52 family of proteins, playing an essential role in the homologous recombination repair (HRR) pathway that is responsible for repairing DNA double-strand breaks and maintaining genomic stability. Structurally, Rad51D forms part of the BCDX2 complex alongside Rad51B, Rad51C, and XRCC2, which is crucial for the detection and repair of DNA damage by facilitating the formation and stabilization of RAD51 nucleoprotein filaments necessary for effective homologous recombination. Additionally, Rad51D interacts with other Rad52 family members like Rad52 and Rad54, enhancing the coordination and efficiency of the DNA repair machinery. Rated 4 out of 5 by from OkWB performed using free sample. Bands correspond to the expected molecular weight. We do not know yet if unspecific bands would appear at higher MW. Western Blot analysis of Rad51D expression on U2-OS and SAOS-2 cells (whole cell lysates, 45ug). The antibody dilution used was 1:500 in the following buffer: Tris-HCl pH 7.5 50 mM, NaCl 50 mM, Tween 0.15%, Skim milk 5%. After O/N incubation at 4°C, the membrane was washed twice in Tris-HCl pH 7.5 50 mM, NaCl 50 mM, Tween 0.15% and once in Tris-HCl pH 7.5 100mM, NaCl 400mM. Incubation was performed 1h at room temperature with anti-mouse secondary antibody 1:5000 (Jackson ImmunoResearch) in 3% Skim milk, Tris-HCl pH 7.5 100mM, NaCl 400mM. The membrane was then washed and chemiluminescent revealed using WESTAR NOVA 2.0 (Cyanagen).
