Rad51 Antibody (F-11) is a mouse monoclonal IgG1 kappa light chain antibody engineered to recognize the C-terminal epitope encompassing amino acids 311-339 of the human Rad51 protein, ensuring precise detection across multiple applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) in mouse, rat, and human samples. Rad51, also called Reca and Rad51a, is a crucial recombinase enzyme involved in the homologous recombination repair (HRR) pathway, essential for the accurate repair of DNA double-strand breaks and maintenance of genomic stability. Structurally, Rad51 assembles into helical nucleoprotein filaments analogous to bacterial RecA protein, enabling critical functions such as searching for homologous DNA sequences and facilitating strand invasion and exchange during repair. The C-terminal region of Rad51 plays a vital role in mediating interactions with other key proteins, including BRCA2 and RAD54, which are integral to DNA damage response and repair mechanisms. These structural domains are essential for Rad51′s ability to form stable filaments, regulate activity, and coordinate with other proteins to ensure efficient and accurate DNA repair, thereby preventing genomic instability, mutations, and potential cancer development. anti-Rad51 antibody (F-11) is available in both non-conjugated form and various conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rad51 (F-11) antibody is recommended for researchers studying molecular mechanisms underlying DNA repair, understanding structural-functional relationships that maintain genomic integrity, and advancing research into cancer development and potential therapeutic interventions.

Hello, I am thinking of trying sc-398587 Rad51 F-11 for IF in human cancer cells but I see some bad reviews for IF (and WB). Do you have protocol recommendations (as far as pre-extracting or not etc)? Thank you.

Thank you for your question. sc-398587 is a great antibody for Rad51. It is recommended and warrantied for IF on human, mouse, and rat samples. Here is a recent publication using it for IF on human samples (PMID: 29694959). Here is a link to the recommended protocol. Do you have the one which can be used for the flow cytometry?

What is the recommended antigen retrieval method when doing IHC(P) with this antibody?

Rated 1 out of 5 by from Didn't work in my hands.Tried for western at 1:500 dilution and didn't detect any band in human neuroblastoma cancer cells. The discontinued works perfectly fine in the same lysates.

Rated 1 out of 5 by from Tried twice for WB, just got a smeary mess.Tried this as a sample, very disappointed. I firstly tried 1:100 5% milk in TBST, incubated for 1.5 hours at room temperature. I got nothing but smears on the blot. I then tried 1:500 incubation for 1.5 hours at room temperature followed by overnight incubation at 4 oC. This is the blot shown in he picture. Nothing at 37kDa and lots of messy smears, does not work at all. Loaded 30ug protein per well with llamaeli buffer. Normally works very well. GAPDH control (not shown) also worked well.
