SEE ALSO...

Pyrin Antibody (C-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects Pyrin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pyrin (C-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pyrin, also known as Marenostrin or Mediterranean fever protein, plays a crucial role in regulating inflammatory response within myelomonocytic cells, particularly influencing cytoskeletal organization. Pyrin′s function is vital in innate immune response, where Pyrin acts as a sensor for cellular stress and infection, leading to activation of inflammatory pathways. Defects in Pyrin gene can result in Mediterranean fever, a hereditary autosomal recessive disorder characterized by recurrent fever and serosal inflammation, highlighting Pyrin′s critical role in maintaining immune homeostasis. Pyrin is predominantly expressed in peripheral blood leukocytes, especially in mature granulocytes, and is absent in lymphocytes, with additional expression noted in spleen, muscle, lung, and various myeloid leukemic, colon cancer, and prostate cancer cell lines.

How much sample do you recommend loading on the gel for Western blot detection using Pyrin (C-11): sc-390938?

Asked by: Randy McDonald
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
