PUMAα Antibody (B-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects PUMA alpha protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PUMAα (B-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PUMA protein, also known as Bcl-2 binding component 3, plays a critical role in regulating apoptosis, particularly in response to cellular stressors such as γ-radiation and glucocorticoids. PUMA is a mitochondrial pro-apoptotic Bcl-2 homology domain (BH3)-only protein that initiates apoptosis through a Bax- and mitochondria-dependent pathway, making PUMA essential for maintaining cellular homeostasis and preventing tumorigenesis. PUMA gene encodes several splice variants, including PUMAα, β, γ, and δ, with PUMAα and PUMAβ containing the crucial BH3 domain that facilitates interaction with anti-apoptotic proteins like Bcl-2 and Bcl-xL. This interaction is vital for the pro-apoptotic function of PUMA, as PUMA helps transmit death signals to the mitochondria, thereby promoting cell death in lymphoid cells. Understanding PUMA function and regulation is important for developing therapeutic strategies targeting apoptosis in cancer and other diseases.

I don't find any signal when i used this antibody for prostate cancer and pencreas. I used 2 methods of Antigen retrieval PH 6/8
