    Protein S Antibody (C-5) is a mouse monoclonal IgG2a antibody that detects Protein S in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Protein S is a crucial vitamin K-dependent plasma protein that plays a significant role in the regulation of blood coagulation and the clearance of apoptotic cells. Anti-Protein S antibody (C-5) functions primarily as a cofactor for activated protein C (APC), enhancing the anticoagulant pathway and preventing excessive clot formation. Notably, Protein S exists in a complex with C4b-binding protein (C4BP) in the plasma, which renders a significant portion inactive; approximately 60-70% of circulating Protein S is bound to C4BP. This calcium-dependent interaction not only influences the regulation of complement activation but also highlights the importance of Protein S in maintaining hemostatic balance. Furthermore, Protein S exhibits APC-independent anticoagulant activity by directly inhibiting prothrombin activation through interactions with Factor Xa, Factor Va, and phospholipids, underscoring its multifaceted role in hemostasis. Deficiencies in Protein S, particularly autosomal dominant forms that result in levels between 15 to 37% of normal, are associated with severe recurrent venous thrombosis, emphasizing the clinical significance of this protein in thrombotic disorders. Protein S monoclonal antibody (C-5) is an essential tool for researchers studying coagulation pathways and related pathologies.
