Protein S Antibody (F-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Protein S protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Protein S antibody (F-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Protein S (PROS) is a crucial vitamin K-dependent plasma protein that plays a significant role in regulating hemostasis by acting as a cofactor for activated protein C (APC), which is essential for inactivating coagulation factors Va and VIIIa, thereby preventing excessive blood clotting. PROS extends beyond anticoagulant properties by facilitating clearance of early apoptotic cells, which maintains tissue homeostasis and prevents autoimmune responses. In plasma, a significant portion of PROS circulates in an inactive form complexed with C4b-binding protein (C4BP), with 60-70% of PROS in this complex. The calcium-dependent interaction between C4BP-PROS and apoptotic cells regulates complement activation, highlighting PROS′s multifaceted roles in both coagulation and immune responses. Notably, autosomal dominant PROS deficiency, characterized by levels ranging from 15 to 37% of normal, associates with heightened risk of severe recurrent venous thrombosis, underscoring PROS′s clinical significance in thrombotic disorders.
