PROC encodes protein C, a vitamin K–dependent serine protease zymogen synthesized in the liver that circulates as a key regulator of hemostasis. Upon activation on the endothelial surface by the thrombin–thrombomodulin complex, activated protein C proteolytically inactivates coagulation cofactors Va and VIIIa, thereby limiting thrombin generation. This anticoagulant pathway is coordinated with endothelial protein C receptor (EPCR) and protein S, linking coagulation control to vascular inflammation and barrier function. Altered PROC expression or function is associated with dysregulated coagulation homeostasis and thrombotic disease phenotypes, supporting its use in mechanistic studies of coagulation signaling and endothelial biology.
Protein C CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PROC gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PROC together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PROC open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Protein C protein expression.
This CRISPR knockout system enables efficient generation of PROC-deficient cell models for investigation of Protein C signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
