PREPL Antibody (E-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects PREPL protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PREPL (E-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PREPL, or Prolyl endopeptidase-like, is a 727 amino acid cytosolic protein that plays a crucial role in regulating peptide metabolism and is part of the prolyl oligopeptidase subfamily of serine peptidases. PREPL functions as a homodimer and exhibits widespread expression, with highest levels found in heart, brain, kidney, and skeletal muscle. A unique mechanism of action, which relies solely on the carboxy-terminal domain for activity, distinguishes PREPL from other family members like prolyl endopeptidase and oligopeptidase B, which require both amino and carboxy-terminal sequences. Defects in the gene encoding PREPL are linked to hypotonia-cystinuria syndrome, also known as 2p21 deletion syndrome, characterized by hypotonia at birth, growth hormone deficiency, and failure to thrive due to homozygous deletions affecting both SLC3A1 and PREPL genes on chromosome 2p21. Alternative splicing of PREPL gene results in production of four distinct isoforms, further underscoring PREPL′s complexity and significance in human health.
