SEE ALSO...

PRAME Antibody (H-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects PRAME protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PRAME antibody (H-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The preferentially expressed antigen of melanoma (PRAME) plays a crucial role in tumor immunology, as PRAME is a tumor-associated antigen primarily expressed in cancer cells, with limited expression in normal tissues, making PRAME a promising target for immunotherapy. PRAME is recognized by cytotoxic T lymphocytes and is expressed in a wide range of malignancies, including melanoma, non-small-cell lung carcinoma, sarcomas, head and neck tumors, and renal carcinomas. High PRAME expression in a significant fraction of tumors underscores PRAME′s potential as a therapeutic target, particularly in melanoma, where PRAME is commonly found. PRAME′s ability to elicit an immune response highlights PRAME′s importance in the development of targeted cancer therapies, as PRAME may help to harness the body′s immune system to combat tumor growth effectively.

How much sample do you recommend loading on the gel for Western blot detection using PRAME (H-10): sc-137188?

Asked by: Professor Griffin
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting

Rated 4 out of 5 by from Works very well in WBWorks very well in WB! Detects a specific signal at approx. 55kDa. The ab has to be used at very high concentrations - for a 'good to detect' signal use 1:100 in 5% milk powder/PBST, when 20 microg of whole protein lysate was loaded/blotted.
