PPOX Antibody (C-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects PPOX protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PPOX (C-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Protoporphyrinogen oxidase (PPOX) plays a crucial role as the penultimate enzyme in the heme biosynthetic pathway, catalyzing the six-electron oxidation of protoporphyrinogen IX to produce protoporphyrin IX, a vital precursor for heme synthesis. Proper functioning of PPOX is essential for maintaining adequate heme levels, which are critical for various biological processes, including oxygen transport and electron transfer in cellular respiration. PPOX protein is localized to the inner membrane of mitochondria across various tissues, such as the heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas, highlighting PPOX importance in energy metabolism and cellular function. Genetic deficiencies in PPOX can lead to variegate porphyria, an autosomal dominant disorder that manifests with cutaneous photosensitivity and neurological symptoms. In rare cases, homozygous variants of this condition result in severe PPOX deficiency, leading to early childhood photosensitization, skeletal abnormalities, and potential cognitive impairments, underscoring PPOX critical role in human health.

What is the recommended fixation method for immunofluorescence staining with PPOX (C-12): sc-271768 monoclonal antibody?

Asked by: Randy McDonald
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
