PP2Cα/β Antibody (D-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects PP2C alpha / beta protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PP2Cα/β (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PP2Cα and PP2Cβ proteins are crucial components of the serine/threonine protein phosphatase family, which plays a significant role in regulating cellular functions through the dephosphorylation of serine and threonine residues. Specifically, PP2Cα is known for dephosphorylating and negatively regulating the activities of mitogen-activated protein kinases (MAPKs) and their upstream kinases, thereby modulating stress response pathways. This regulation is vital for maintaining cellular homeostasis and preventing uncontrolled cell proliferation, as PP2Cα also promotes the expression of p53 and its target gene p21, leading to cell cycle arrest and apoptosis in response to stress. PP2Cα features a dinuclear metal ion center in its active site, which is essential for phosphatase activity, and is predominantly expressed in epithelial cells, as well as in organs such as the digestive tract, lung, kidney, breast, prostate, endocrine glands, and brain. PP2Cβ, which is also broadly specific, is highly expressed in cardiac and skeletal muscle tissues and has been implicated in cell cycle control by dephosphorylating cyclin-dependent kinases (CDKs) such as CDK2 and CDK6, with overexpression potentially leading to cell growth arrest or apoptosis.
