POU2F3 CRISPR/Cas9 KO Plasmid (h) | sc-417495 | 20 µg | $397.00 |
POU2F3 encodes a POU domain transcription factor that regulates lineage-specific gene expression programs in epithelial cells, with prominent roles in differentiation and maintenance of specialized tuft/brush cell states. By binding octamer-like motifs and coordinating transcriptional networks, POU2F3 influences epithelial identity, sensory-associated signaling, and mucosal barrier-associated processes. Altered POU2F3 activity has been linked to lineage plasticity and subtype-specific transcriptional dependencies in certain epithelial-derived cancers, including small cell lung cancer variants. As a master regulator, it is frequently studied in the context of chromatin-driven cell fate control and transcription factor circuitry rewiring.
POU2F3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the POU2F3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the POU2F3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the POU2F3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish POU2F3 protein expression.
This CRISPR knockout system enables efficient generation of POU2F3-deficient cell models for investigation of POU2F3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
