    Overview

    POMC (proopiomelanocortin) encodes a secretory prohormone that is proteolytically processed into multiple bioactive peptides, including ACTH and melanocyte-stimulating hormones, which coordinate endocrine stress signaling, energy homeostasis, pigmentation, and nociception. In pituitary corticotrophs, POMC-derived ACTH drives adrenal steroidogenesis through melanocortin receptor pathways, linking hypothalamic–pituitary–adrenal axis activity to systemic metabolic and immune responses. POMC processing depends on regulated secretory pathway trafficking and prohormone convertases, integrating with cAMP/PKA signaling and neuroendocrine differentiation programs. Dysregulation or loss of POMC function is associated with disorders of adrenal function, obesity and hyperphagia phenotypes, and altered melanocortin signaling relevant to neuroendocrine and metabolic research.

    POMC CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the POMC gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the POMC together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the POMC open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish POMC protein expression.

    This CRISPR knockout system enables efficient generation of POMC-deficient cell models for investigation of POMC signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
