POLA2 Antibody (D-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects POLA2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). POLA2 (D-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. POLA2 protein, also known as DNA polymerase subunit alpha B, is a crucial component of the DNA polymerase alpha enzyme complex, which consists of four subunits: the large and small subunits of DNA primase, along with DNA polymerase subunits A and B. This complex plays a vital role in DNA replication, particularly during the early stages when coupling the primase/polymerase complex to the replication machinery, ensuring accurate and efficient DNA synthesis. POLA2 is composed of 598 amino acids, and incorporation into the enzyme complex is facilitated by its 250 amino acid N-terminal domain. Nuclear localization of POLA2 is essential for function, and phosphorylation may occur during the G2/M phase of the cell cycle, a modification that could regulate activity and interactions with other proteins involved in DNA replication.

You say the second band is due to the fact there are two isoforms. One isoform is 66 KDa, the other around 40 KDa. I would expect a band running faster in the gel. Right?

Thank you for your question. DNA polymerase alpha subunit B has a truncated alterntive splicing isoform that contains the epitope sequence the antibody targets, which is why the antibody detects both isoforms in western blots.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with POLA2 (D-2): sc-398255?

We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
