    PNPO Antibody (E-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects PNPO protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-PNPO antibody (E-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PNPO, also known as pyridoxamine 5′-phosphate oxidase, is a crucial enzyme composed of 261 amino acids that plays a vital role in vitamin B6 metabolism. Specifically, PNPO catalyzes the conversion of pyridoxine 5′-phosphate (PNP) and pyridoxamine 5′-phosphate (PMP) into pyridoxal 5′-phosphate (PLP), the active form of vitamin B6, which is essential for numerous enzymatic reactions in amino acid metabolism and neurotransmitter synthesis. Proper functioning of PNPO is critical, as deficiencies can lead to serious neurological disorders, including PNPO-related neonatal epileptic encephalopathy and potential links to schizophrenia. Encoded by a gene located on human chromosome 17q21.32, PNPO is characterized by a structure that includes seven exons and six introns, with all exon/intron junctions adhering to the GT/AG consensus splicing site. PNPO′s regulatory region features Sp1-binding sites and CpG islands, indicative of its role as a housekeeping gene, while PNPO expression is developmentally regulated in tissues such as the liver, brain, skeletal muscle, and kidney, with minimal expression in the lung.

    Rated 5 out of 5 by from Excellent product!I tested the antibody in the wild type and PNPO knock out Hek293 total cell lysates and it works really well! A single clear band is visible at the expected MW in the wild type cell lysates, no signal in the knock outs. I loaded 20ug/well protein and used 1:1000 dilution of the antibody.
