PNPase Antibody (A-4) is a mouse monoclonal IgG2b antibody that detects polyribonucleotide nucleotidyltransferase 1 (PNPase) in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PNPase plays a crucial role in mRNA degradation, hydrolyzing single-stranded ribonucleotides in the 3′ to 5′ direction, which is essential for regulating gene expression and maintaining cellular homeostasis. PNPase is primarily located in the mitochondria, where PNPase is involved in the processing and degradation of mitochondrial RNA, thereby influencing mitochondrial function and energy production. Notably, PNPase forms homotrimers and is upregulated in response to interferon-β induction, highlighting PNPase′s role in cellular stress responses. The N-terminus of PNPase contains a putative mitochondrial targeting sequence, and mutation analysis has confirmed that these sequences are critical for directing PNPase to the mitochondria, ensuring proper localization and function. Endogenous PNPase has been shown to co-localize with mitochondrial marker proteins in HeLa cells, further emphasizing PNPase′s importance in mitochondrial dynamics and RNA metabolism.
