PMCA4b Antibody (JA3) is a mouse monoclonal IgG1 kappa light chain antibody that detects PMCA4b protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-PMCA4b antibody (JA3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The plasma membrane Ca2+-pumping ATPase (PMCA) family consists of four genes, PMCA1-4, which undergo alternative splicing to produce "b" splice forms that contain PDZ interaction domains, allowing them to interact with different members of the PSD95 family. PMCA4b is predominantly expressed in developing mammary tissue, where PMCA4b plays a crucial role in regulating calcium homeostasis during lactation. The expression of PMCA1b increases while PMCA4b decreases during this period, highlighting the dynamic regulation of calcium transport in response to physiological demands. PMCA4b is cleaved by caspase-3 at Asp 1080, resulting in a fully active fragment that responds more rapidly to calcium fluctuations than its autoinhibited counterpart. Anti-PMCA4b antibody (JA3) is essential for studying PMCA4b′s role in maintaining low cytosolic calcium levels in resting platelets and potential involvement in apoptotic processes, where PMCA4b may help regulate intracellular calcium levels.
