PLCB3 encodes phospholipase C beta 3 (PLC β3), a membrane-associated effector downstream of GPCR and certain receptor tyrosine kinase signals that hydrolyzes PIP2 to generate the second messengers IP3 and DAG. This reaction drives intracellular Ca2+ mobilization and protein kinase C activation, shaping signal transduction programs that regulate proliferation, migration, secretion, and immune cell function. PLC β3 activity integrates with calcium signaling, MAPK cascades, and cytoskeletal remodeling, linking extracellular cues to transcriptional and metabolic responses. Dysregulated PLCB3-associated signaling has been implicated in oncogenic and inflammatory contexts, making it a useful node for dissecting pathway wiring in disease-relevant cellular models.
PLC β3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PLCB3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PLCB3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PLCB3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PLC β3 protein expression.
