SEE ALSO...

PKA IIα reg Antibody (H-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects PKA II alpha reg protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PKA IIα reg (H-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PKA II alpha reg protein, also called protein kinase cAMP-dependent type II regulatory subunit alpha, PRKAR2A, PKR2, or RIIα, plays a crucial role in cellular signaling pathways by acting as a regulatory subunit of the protein kinase A (PKA) complex, which is activated by the second messenger cyclic AMP (cAMP). Upon binding of cAMP to regulatory subunits, active catalytic subunits are released, leading to phosphorylation of various target proteins, including transcription factors. This phosphorylation is essential for mediating diverse cellular responses such as gene transcription, metabolism regulation, and cell proliferation. PKA II alpha reg interacts with specific proteins, including the cAMP response element binding protein (CREB), which is phosphorylated at serine 133, facilitating recruitment of transcriptional machinery to initiate gene expression. PKA′s ability to translocate to the nucleus and influence transcription underscores PKA II alpha reg′s importance in cellular responses to external signals, making anti-PKA IIα reg antibody (H-12) a valuable tool for studying these critical signaling pathways in human cells.

Hello, Am I able to detect PKA in Rat brain with this antibody (PKA IIα reg Antibody (H-12): sc-137220) in WB?

Asked by: Jasem
Thank you for your question. We do not currently recommend using this antibody, PKA IIα reg (H-12): sc-137220 , for detection of PKA IIα reg in rat samples. This antibody is currently only recommended for human. As an alternative, we would recommend using our PKA IIα reg Antibody (40): sc-136262, which is recommended for mouse, rat and human samples. https://www.scbt.com/scbt/product/pka-iialpha-reg-antibody-40

I am using PKA IIα reg (H-12): sc-137220 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: DefinitelyNotMatt
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
