PI 3-kinase p85α/β/p55γ Antibody (D-9) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 323-596 within an internal region of human PI 3-kinase p85β, with reactivity in mouse, rat, and human species. PI 3-kinase p85α/β/p55γ (D-9) antibody is validated for western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PI 3-kinase, also known as phosphoinositide-3-kinase, is a heterodimer composed of a regulatory subunit (p85) and a catalytic subunit (p110), and phosphorylates phosphoinositides at the 3′ OH position of their inositol rings. The p85 subunits, including p85α, p85β, and p55γ, serve as regulatory adapters that anchor the catalytic subunit to activated receptor tyrosine kinases at the plasma membrane. Such localization is essential for signal transduction, as positioning near lipid substrates initiates pathways like AKT/mTOR that regulate cell growth, survival, and metabolism. PI 3-kinase p85α (also known as GRB1 or phosphatidylinositol 3-kinase regulatory 1) plays a key role in insulin signaling, and dysregulation has been linked to conditions such as insulin resistance and cancer. Post-translational modifications, including tyrosine phosphorylation and ubiquitination, modulate p85 subunit activity and stability, further refining signaling pathway control. Anti-PI 3-kinase p85α/β/p55γ antibody (D-9) serves as a valuable tool for studying cellular localization and regulatory roles of PI 3-kinase subunits in signaling and disease.

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PI 3-kinase p85α/β/p55γ (D-9): sc-374534 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

There was strong detection of the ~84/53/50/84 kDa target proteins in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested.
