PHI-1 Antibody (D-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects PHI-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PHI-1 (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PHI-1, also known as PPP1R14B (protein phosphatase 1, regulatory (inhibitor) subunit 14B), PLCB3N (phospholipase C-beta-3 neighboring gene protein), PNG, or SOM172, is a 147 amino acid coiled-coil protein that plays a crucial role in cellular signaling by inhibiting protein phosphatase 1 (PP1). This inhibition is vital as PP1 is involved in various cellular processes, including cell division, metabolism, and muscle contraction. PHI-1 is primarily located in the cytoplasm and is expressed at low levels across different tissues, where PHI-1 functions to block the dephosphorylation of myosin light chains, thereby influencing cell migration and adhesion. Notably, PHI-1 interacts with MYPT1, a significant regulator that may act as a tumor suppressor by modulating Rho-dependent amoeboid cell behavior during metastasis. PHI-1 undergoes post-translational modifications, particularly phosphorylation on threonine 57 and an unidentified serine by protein kinase C, which enhances inhibitory activity by 50-fold, underscoring the importance of these modifications in regulating function. The gene encoding PHI-1 is located on human chromosome 11q13.1, highlighting relevance in both normal physiology and potential pathological conditions.
