PGM 1 Antibody (Y-173) is a mouse monoclonal IgG1 antibody that detects phosphoglucomutase 1 (PGM1) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PGM1 antibody (Y-173) plays a crucial role in the regulation of carbohydrate metabolism, specifically in glycogen catabolism and synthesis. PGM1 is responsible for the interconversion of glucose-1-phosphate and glucose-6-phosphate, which is vital for maintaining glucose homeostasis in the body. PGM1′s activity is essential for glycolysis and the pentose phosphate pathway, both of which are critical for energy production and cellular function. Notably, PGM1 exists as a highly polymorphic protein with three genetic loci—PGM1, PGM2, and PGM3—each encoding isozymes that catalyze the same biochemical reactions but differ in their amino acid sequences. PGM1 isozyme, consisting of 562 amino acids, exhibits significant genetic diversity due to mutations and recombination events, resulting in eight distinct protein variants. Each phosphoglucomutase functions as a monomer and binds a single magnesium ion per subunit, which is essential for enzymatic activity. PGM1 monoclonal antibody (Y-173) is an invaluable tool for researchers studying the intricate mechanisms of carbohydrate metabolism and its implications in various metabolic disorders.

I am using PGM 1 (Y-173): sc-100411 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Randy McDonald
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
