Phosphofructosekinase-2 (PFK-2) plays a crucial role in cellular metabolism by regulating fructose 2,6-bisphosphate (F2,6BP) levels, a key metabolite that activates 6-phosphofructo-1-kinase, promoting glycolysis. PFK-2′s ability to catalyze fructose-6-phosphate phosphorylation to produce F2,6BP maintains balance between glycolytic and gluconeogenic pathways under varying physiological conditions. PFK-2 exists in several tissue-specific isoforms, including brain, liver, and placenta variants, each differentially regulated and functioning as a homodimer. iPFK-2, a unique isoform, is induced by proinflammatory stimuli and constitutively expressed in various carcinoma cell lines, leading to intracellular F2,6BP accumulation. This accumulation associates with increased DNA synthesis, suggesting iPFK-2′s role in cellular transformation and enhanced proliferation, making iPFK-2 a potential target for therapeutic intervention in cancer and metabolic disorders.
