    PFAAP5 Antibody (F-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects PFAAP5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PFAAP5 (F-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PFAAP5 protein, also known as phosphonoformate immuno-associated protein 5 or NEDD4-binding protein 2-like 2, is a 583 amino acid nuclear protein that plays a crucial role in transcriptional regulation, particularly in response to DNA damage. PFAAP5 is phosphorylated on serine 199, likely by ATM or ATR kinases, which is essential for function in cellular stress responses. Primarily expressed in bone marrow, PFAAP5 is significantly down-regulated following exposure to arsenic compounds, a change that precedes the onset of neutropenia, highlighting importance in hematopoiesis and immune response. PFAAP5 interacts with Gfi-1 and Neutrophil Elastase, both implicated in neutropenia disorders. Mutations in the gene encoding Neutrophil Elastase, ELA2, lead to cyclic hematopoiesis, characterized by reduced neutrophil counts and increased susceptibility to opportunistic infections. Gfi1 acts as a transcriptional repressor targeting the ELA2 gene, and PFAAP5 can enhance the repression of Gfi1 target genes, facilitating the transcription of ELA2 and promoting neutrophil differentiation. Two isoforms of PFAAP5 arise from alternative splicing, further diversifying functional roles in cellular processes.
