Periostin Antibody (F-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Periostin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Periostin antibody (F-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Periostin, also known as osteoblast-specific factor 2 (OSF-2), is a disulfide-linked secreted protein that plays a crucial role in cell adhesion and migration, particularly in bone and tissue remodeling. Periostin serves as a ligand for alpha(V)beta(3) and alpha(V)beta(5) integrins, facilitating osteoblast recruitment and attachment, essential for bone formation and repair. Periostin is predominantly found in the lower gastrointestinal tract, aorta, stomach, placenta, uterus, and breast tissues, with notable up-regulation in epithelial ovarian tumors and overexpression in breast cancer, highlighting potential roles in tumor progression and metastasis. Periostin expression is influenced by bone morphogenetic protein 2 (BMP2) and transforming growth factor beta 1 (TGF β 1), indicating involvement in various signaling pathways that regulate cellular processes. Structurally, Periostin features a typical signal sequence, a cysteine-rich domain, a fourfold repeated domain homologous to the insect protein fasciclin, and a C-terminal domain, which together contribute to functional properties and interactions within the extracellular matrix.

Hello. I used this antibody for western blot. However, the periostin was detected at about 70kda. Is it possible? Cell lines were HepG2 and LX-2.

Thank you for your question. Periostin has been reported to be expressed as many different isoforms. The full length protein is often described in the literature as a 90 kDa secreted glycoprotein. The literature has also described isoforms at ~84 kDa and 74 kDa with cleavages in the C-terminal region (CTR), see PMID: 19887451 as a reference.

Can Periostin (F-10): sc-398631 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Periostin (F-10): sc-398631 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
