SEE ALSO...

PDK2 Antibody (S-15) is a mouse monoclonal IgG2a kappa light chain antibody that detects PDK2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-PDK2 antibody (S-15) is available as the non-conjugated form. PDK2, a member of the pyruvate dehydrogenase kinase family, plays a crucial role in cellular metabolism by regulating the activity of the pyruvate dehydrogenase complex (PDC) through phosphorylation of its E1α subunit. This regulation is vital as it determines whether pyruvate is converted into acetyl-CoA for energy production or diverted towards other metabolic pathways, particularly during periods of nutrient deprivation. The PDC is composed of multiple dihydrolipoamide acetyltransferase (E2) subunits, which interact with PDK2 to enhance its kinase activity, thereby influencing metabolic flexibility. Notably, PDK2 expression is upregulated under starvation conditions, promoting fatty acid oxidation and altering acetyl-CoA generation to meet the energy demands of the cell. PDK2′s structure includes five conserved regions that are essential for kinase activity, and PDK2 shares mechanistic similarities with bacterial His-kinases, highlighting evolutionary significance. PDK2 transcripts are widely expressed across various tissues, with particularly high levels found in heart and skeletal muscle, underscoring importance in energy metabolism in these tissues.

Can sc-100534: PDK2 (S-15) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: AbPolly
Thank you for your question. Yes, sc-100534: PDK2 (S-15) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
