PCMT1 Antibody (D-E7) is a mouse monoclonal IgG1 kappa light chain antibody that detects PCMT1 protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). Anti-PCMT1 antibody (D-E7) is available as a non-conjugated format. PCMT1 protein, also known as protein-L-isoaspartate (D-aspartate) O-methyltransferase or PIMT, plays a crucial role in repairing and degrading damaged proteins, vital for maintaining cellular health and function. PCMT1 is predominantly expressed in brain tissue and operates as a monomer localized in the cytoplasm. PCMT1 specifically recognizes isomerized aspartate or asparagine residues in proteins and catalyzes their conversion to methyl esters, which can subsequently hydrolyze to restore normal aspartyl residues. This enzymatic activity prevents accumulation of damaged proteins that can lead to cellular dysfunction. Studies in knockout mice show that PCMT1 deficiency results in significant buildup of damaged proteins across various tissues, increased ratio of S-adenosylmethionine to S-adenosylhomocysteine in brain tissue, ultimately leading to severe phenotypic consequences such as progressive epilepsy and early mortality.
