    Pax-4 Antibody (3C12) is a mouse monoclonal IgG1 antibody that detects Pax-4 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Pax-4 (3C12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pax-4 protein, also known as paired box gene 4, plays a crucial role in the differentiation of insulin-producing beta cells and is essential for normal pancreatic islet development. Located primarily in pancreatic islets, Pax-4 functions as a transcriptional repressor, binding to a common cis element in promoters of glucagon, insulin, and somatostatin, thereby regulating their expression. Pax-4 presence in pancreatic islets and specific β cell lines, such as MIN6, β TC, and NIT1, highlights its importance in endocrine pancreas differentiation, where Pax-4 may compete with Pax-6 for binding sites, thus influencing the transcriptional landscape of these critical hormones. Human Pax-4 gene encodes a deduced protein of 350 amino acids, exhibiting 80% identity with its mouse counterpart, underscoring the evolutionary conservation of its function in glucose homeostasis and metabolic regulation.

    How much sample do you recommend loading on the gel for Western blot detection using Pax-4 (3C12): sc-517254?

    We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.
