PAH Antibody (H-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects PAH protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PAH Antibody (H-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The PAH gene encodes phenylalanine hydroxylase, which plays a crucial role in converting phenylalanine to tyrosine, serving as the rate-limiting enzyme in phenylalanine catabolism. This enzymatic function is vital for proper metabolism of amino acids and prevention of toxic phenylalanine accumulation, which can lead to serious neurological issues. PAH protein is primarily located in the liver, where abundant expression highlights this organ′s central role in amino acid metabolism. Deficiency in PAH activity leads to phenylketonuria (PKU), an autosomal recessive disorder characterized by cognitive impairment if not managed with a low-phenylalanine diet from an early age. The PAH gene, located on human chromosome 12q23.2, contains all necessary genetic information to produce functional PAH, illustrating the significance of this single gene in the classic disease phenotype. Various mutations in PAH can result in decreased enzyme activity, leading to different severities of PKU, and while multiple isozymes of PAH have been reported, these are likely allelic variants that produce protein subunits with slightly different charges and electrophoretic migration patterns.

Dear SCBT team. I need an anti PAH antibody for FLOW and WB that don't have mouse reactivity, that can stain human PAH positive cells in mouse samples. Many thanks

Asked by: Dima Yackoubov
Thank you for your question. We currently do not have any antibodies specific against this target that do not also recognize murine protein.

What application is the blocking peptide sc-271258 P appropriate for?

Asked by: cjMara
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
