p54/nrb Antibody (3) is a mouse monoclonal IgG1 antibody that detects p54/nrb in mouse, rat, human, and canine samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). p54/nrb, a crucial nuclear RNA-binding protein, plays a significant role in the formation and function of paraspeckles, which are essential for the regulation of gene expression and RNA processing. These discrete bodies are located in the interchromatin nucleoplasmic space and are involved in the dynamic movement of RNA-binding proteins, including p54/nrb, between the nucleolus and paraspeckles. The presence of p54/nrb in paraspeckles is vital for the proper accumulation of splicing factors, as p54/nrb co-localizes with splicing speckles, thereby influencing pre-mRNA splicing pathways. p54/nrb contains two RNA recognition motifs and shares extensive homology with the human splicing factor PSF and Drosophila NONA/BJ6, which is essential for normal vision and courtship behaviors. The shared DBHS (Drosophila behavior, human splicing) domain among these proteins suggests a conserved mechanism that may regulate various pathways at the level of pre-mRNA splicing, highlighting the importance of p54/nrb in cellular processes and its potential implications in understanding splicing-related diseases.

I am using p54/nrb (3): sc-136296 for immunofluorescence, what dilution of this antibody should I use?

Asked by: DefinitelyNotMatt
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
