Hi! Does this antibody recognize p25, the proteolytic product of p35?

Thank you for your question. According to Uniprot, (http://www.uniprot.org/uniprot/Q15078#ptm_processing), the cleaved p25 form of p35 corresponds to amino acids 99-307. This mouse monoclonal antibody, p35 (4G11): sc-293184, is raised against amino acids 208-307 of human p35. Therefore, the antibody does have an epitope that should recognize the cleaved p25 form. We have not confirmed this experimentally.

I am using p35 (4G11): sc-293184 for immunofluorescence, what dilution of this antibody should I use?

WB 1:1000 Overexpressed p35 in 293 cells or endogenous p35 from embryonic mouse brain (cortex) lysates.

Rated 4 out of 5 by from works good for western blotI got the free sample the last week and I got clear bands for western blot using lysates from cultured neurons and mouse brains. In addition, It successfully detects p35 and p25 in cell lysates. My PVDF membrane is probed with this antibody in 3% milk/TBST with 1:500 dilution at 4oC for 1 overnight.

Rated 1 out of 5 by from This p35 (4G11) antibody didn't work on WB.I got the p35 (4G11) antibody a few days ago and used the recommended secondary antibody sc-516102 to get the signal of LNCaP protein lysate by western blotting. There is nothing. No band (signal) on PVDF membrane. =>Primary antibody dilution is 1:1500~1:750, tried and got nothing. =>Plus secondary antibody dilution from 1:10000 to 1:5000, also got nothing. I got normal signal one the same target and same cell lysate and same WB method, the p35 (c-19) sc-820 works fine and perfect. Very sad!!
