p-Stat4 Antibody (pY693.38) is a mouse monoclonal IgG2b antibody that detects p-Stat4 in mouse, rat, and human samples through applications such as western blotting (WB) and immunoprecipitation (IP). p-Stat4 monoclonal antibody (pY693.38) plays a crucial role in the signaling pathways activated by various ligands, including interferons and growth hormones, which induce the activation of Janus kinases (Jak). This activation leads to the tyrosine phosphorylation of Stat transcription factors, particularly Stat4, which is essential for mediating interleukin-12 (IL-12) signals. Stat4 is predominantly expressed in testis and myeloid cells, and Stat4 activation is vital for the immune response, as p-Stat4 facilitates the transcription of genes involved in T cell differentiation and function. Upon phosphorylation, Stat4 dimerizes and translocates to the nucleus, where p-Stat4 binds to specific regulatory elements that control gene expression. Notably, the phosphorylation of Ser 721 of Stat4 is dependent on p38 MAPK and is critical for transcriptional activity, highlighting the importance of post-translational modifications in regulating Stat4′s function in immune signaling.
