p-Atm Antibody (10H11.E12) is a mouse monoclonal IgG1 kappa light chain antibody that detects Ser 1981 phosphorylated Atm of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. p-Atm (10H11.E12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Atm protein plays a crucial role in cellular response to DNA damage, particularly in detecting and repairing double-strand breaks. This function maintains genomic stability, as mutations in Atm gene are associated with ataxia telangiectasia (AT), leading to severe neurological and immunological defects. In response to DNA damage, Atm rapidly activates through autophosphorylation at serine 1981, initiating a cascade of signaling events that lead to cell cycle arrest and DNA repair. Most human tissues predominantly express non-phosphorylated, inactive Atm, highlighting the importance of post-translational modifications in regulating activity and function. anti-Atm antibody (10H11.E12) specifically recognizes phosphorylated Atm, making p-Atm (10H11.E12) monoclonal antibody an invaluable tool for studying DNA damage response mechanisms and pathophysiology of related disorders.

Dear Santa Cruz. We purchased pATM (sc-47739) and I cannot get the western blots to detect it. Could you please send me info on how to extract the protein (cells/heart tissue), and what WB steps you recommend to get it to work? Very much appreciated.

Hi All, Are these antibodies p-ATM Antibody(10H11.E12):sc-47739(Alexa flour 647)and P53(sc-126 Alexa flour647) suitable to detect ATM and P53 in Bone marrow mouse samples using flow cytometry? Kind regards, Ahmad

What is a good negative control to test for phospho-specificity of this antibody?

Thank you for your question. We recommend treating your samples with lambda protein phosphatase (sc-200312A) as a control. Lambda Phosphatase can be used to release phosphate groups from phosphorylated serine, threonine and tyrosine residues in proteins. As indicated in the Western blot data on our website and datasheet for this antibody (p-Atm Antibody (10H11.E12): sc-47739), treated samples should show a negative result when using this phospho-specific antibody in WB compared to the control phosphorylated sample.
