Oxytocin-R Antibody (C-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Oxytocin-R of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Oxytocin-R antibody (C-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Oxytocin-R, also known as oxytocin receptor, is a critical integral membrane protein belonging to the G protein-coupled receptor family, which plays a vital role in mediating physiological effects of oxytocin, a hormone involved in childbirth and lactation. Oxytocin-R is predominantly located in the uterus and cervix, where expression significantly increases during pregnancy in response to both endocrine and mechanical signals. This localization facilitates uterine contractions necessary for parturition and milk ejection reflex during nursing. Additionally, regulation of Oxytocin-R expression is essential for maintaining reproductive health, as inhibition by factors such as interferon-alpha and interferon-tau may contribute to suppression of oxytocin signaling during early pregnancy, highlighting Oxytocin-R′s importance in reproductive processes.

I would like to find out if you have developed any good antibodies for OTR immunocytochemistry? It appears that Oxytocin-R Antibody (C-4): sc-515809 does not work well for ICC? We are searching for an antibody to use in California mice. Thanks!

Thank you for your question. Unfortunately this is the only antibody we offer for Oxytocin-R at this time. It is recommended for WB, IP, IF and ELISA, but has not been validated for immunohistochemistry yet. IF does include IHC on fresh frozen tissue sections. How much sample do you recommend loading on the gel for Western blot detection using Oxytocin-R (C-4): sc-515809?

Why are there two bands with this anti-OTR antibody? I've never seen that before with any other anti-OTR blot using human uterine tissue. I was re-directed here when searching for the discontinued sc-33209 anti-OTR antibody, which only had a single band.

Thank you for your question. The base predicted weight for Oxytocin-R before any post-translational modification is 42 kDa, and the observed weight following modification such as glycosylation is approximately 66 kDa. This particular sample appears to express both forms at detectable levels. This may not be the case for all samples. Detected Oxytocin at expected size in 15 ug total proteine lysate, wet transfer one hour at 450 mAmps on PVDF membrane, overnight blocking with 5% nonfat dry milk in PBS, one hour incubation at RT with 1:500 anti Oxytocin R in 5% PBS with 0,2%Tween followed by one hour incubation with 1:10.000 anti mouse (sc-358914) HRP conjugated antibody and detection with ECL prime substrate followed by short film exposure (under 1 minute) and automatic processing.
