    Nucleobindin Antibody (D-8) is a mouse monoclonal IgM antibody that detects nucleobindin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-nucleobindin antibody (D-8) is available as the non-conjugated form. Nucleobindin, also known as Nuc or Calnuc, is a secreted protein that plays a crucial role in the immune response, particularly in promoting the production of DNA-specific antibodies in lupus-prone MRL/lpr mice, highlighting its significance in autoimmune disorders. Structurally, nucleobindin features a signal peptide, two EF-hand motifs, acidic and basic regions, and a leucine-zipper motif, which are essential for function. The presence of two calcium-binding domains allows nucleobindin to act as a major Golgi Ca2+ binding protein, facilitating calcium homeostasis within the cell. The leucine zipper structure and the basic amino acid-rich region are critical for nucleobindin′s ability to bind DNA, which is vital for regulating gene expression and cellular responses. Nucleobindin is predominantly localized to the cis-Golgi cisternae and the cis-Golgi network, as well as being found in both the cytosol and the membrane of polarized cells, underscoring its importance in cellular signaling and function. Through involvement in autoimmunity, apoptosis, and calcium regulation in the bone matrix, nucleobindin serves as a key player in maintaining cellular health and responding to stress signals.
