Notch1 Antibody (mN1A) is a mouse monoclonal IgG1 kappa light chain antibody developed to detect Notch1, also known as Neurogenic locus notch homolog protein 1, in mouse, rat, and human samples via western blotting (WB) and immunoprecipitation (IP). Notch1 is a single-pass transmembrane receptor that plays a pivotal role in cell fate determination during embryonic and adult development. Upon binding to its ligands, such as Jagged1, Jagged2, and Delta-like proteins, Notch1 undergoes proteolytic cleavage, releasing its intracellular domain, which translocates to the nucleus to regulate gene expression. This signaling pathway is crucial for processes like differentiation, proliferation, and apoptosis, impacting the development of various tissues and organs. Dysregulation of Notch1 has been implicated in several cancers, including T-cell acute lymphoblastic leukemia, highlighting its importance in maintaining cellular homeostasis. anti-Notch1 antibody (mN1A) is available in non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, catering to diverse experimental needs.

What dilution and detecting agent to use for this antibody? I am having trouble detecting this. Picture in the data sheet looks good.

Thank you for your question. We used the primary antibody, Notch 1 (mN1A): sc-32745, at a dilution of 1:200 for the Western blot data shown on our product data sheet. We used a conventional anti-mouse IgG-HRP conjugated secondary antibody, and our Luminol (sc-2048) for detection.
