  • specific for an epitope mapping between amino acids 2425-2444 near the C-terminus of Notch 1 of human origin
  • recommended for detection of Notch 1 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA
  • available conjugated to agarose for IP; HRP for WB, IHC(P) and ELISA; phycoerythrin, FITC, Alexa Fluor® 488 or Alexa Fluor® 647 for IF, IHC(P) and FCM
  • m-IgGκ BP-HRP is the preferred secondary detection reagent for Notch1 Antibody (A-8) for WB and IHC(P) applications. This reagent is now offered in a bundle with Notch1 Antibody (A-8) (see ordering information below). For additional m-IgGκ BP conjugates see our complete list of Mouse IgG Binding Proteins.

Notch 1 Antibody (A-8) is a mouse monoclonal IgM kappa light chain antibody specifically engineered to detect Notch1, also known as Neurogenic locus notch homolog protein 1, in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. Notch1 monoclonal antibody (A-8) targets an epitope spanning amino acids 2425-2444 near the C-terminus of the Notch1 protein, ensuring precise recognition of the Notch1 precursor, mature Notch1, Notch1 NEXT, and Notch1 NICD forms across multiple species, including equine. Notch1 is a transmembrane receptor that plays a pivotal role in the Notch signaling pathway, which is essential for regulating cell fate decisions, differentiation, proliferation, and apoptosis during embryonic development and maintaining tissue homeostasis in adults. By interacting with ligands such as Jagged1, Jagged2, and Delta-like proteins, Notch1 facilitates critical communication between neighboring cells, influencing various developmental processes and ensuring proper cellular function. These interactions are vital for processes like vascular development, neural differentiation, and immune system regulation. Dysregulation of Notch1 signaling has been implicated in numerous diseases, including various cancers and developmental disorders, highlighting the importance in maintaining cellular balance and preventing pathological conditions. anti-Notch1 antibody (A-8) is available in both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, catering to diverse experimental needs.

Hello, I would like to use Notch-1 Antibody (A-8) primarily for western blot and later maybe for IC. Could you let me know if I could purchase the antibody conjugated to Alexa Fluor 488 and use it for WB Too or for WB only the HRP conjugated works?

Asked by: myrto
Yes, you can use conjugates to Alexa Fluor 488 for Western blotting.

Can I use this antibody for IF-P staining? If not which product can I use for IF tissue staining?

Asked by: JordanA
Thank you for your question. This antibody is recommended for IF and IHC(P), or immunofluorescence and immunohistochemistry on formalin-fixed paraffin-embedded tissue sections.

I purchased this Notch 1 antibody (A-8) to do western blot for human breast cancer cell lines lysates, but I am not sure where the band will appear in the gel (110KD or 210 KD?). Please tell me a relatively accurate position. THX~

Asked by: Tianzi
Thank you for your question. The expected molecular weight for Notch one is ~300 kDa (precursor) and two possible cleaved forms which run around ~180 and ~115-120kDa.
