NOSTRIN Antibody (F-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects NOSTRIN of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). NOSTRIN (F-10) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NOSTRIN, also known as nitric oxide synthase trafficker isoform 1 or endothelial nitric oxide synthase traffic inducer, is a crucial member of the Pombe Cdc15 homology (PCH) family of proteins. NOSTRIN is predominantly expressed in vascular endothelial cells of highly vascularized tissues such as placenta, lung, kidney, and heart, where NOSTRIN plays a vital role in regulating nitric oxide (NO) production. Structurally, NOSTRIN consists of an N-terminal Cdc15 domain, an FCH (Fes/CIP homology) region, two coiled-coil domains, and a C-terminal SH3 domain, typically existing as a trimer. NOSTRIN functions as an adaptor, binding to caveolin-1 through an internal domain and to NOS3 via its SH3 domain, thereby forming a ternary complex that facilitates caveolar transport of NOS3. This transport is critical because NOS3 is responsible for producing nitric oxide, a key mediator in numerous biological processes, including vasodilation and neurotransmission. NOSTRIN-mediated translocation of NOS3 from plasma membrane to intracellular vesicle-like structures can significantly diminish NO production, highlighting NOSTRIN′s importance in maintaining vascular homeostasis and potential implications in cardiovascular diseases. NOSTRIN also interacts with Dynamin and N-WASP through its SH3 domain, further underscoring NOSTRIN′s role in cellular signaling and transport mechanisms.
