NKp46 Antibody (9E2) is a mouse monoclonal IgG1 kappa light chain antibody that detects NKp46 protein of human origin by western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). Anti-NKp46 antibody (9E2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NKp46, also known as lymphocyte antigen 94 (LY94), is a crucial natural cytotoxicity receptor belonging to the immunoglobulin superfamily, predominantly expressed on all resting and activated natural killer (NK) cells, while absent on T and B cells. Structurally, NKp46 is a type I transmembrane protein consisting of 304 amino acids, featuring an extracellular region that is preceded by a 21-residue signal peptide and contains two cysteine-bridged C2-type immunoglobulin-like domains. The importance of NKp46 structure lies in the stem that connects the extracellular domain to a 19-amino acid transmembrane domain rich in arginine, which is essential for mediating natural cytotoxicity. NKp46 plays a pivotal role in the recognition and lysis of both human and murine tumor cells, allowing NK cells to target cells infected by viruses such as influenza or parainfluenza, even in the presence of reduced expression of MHC class I proteins on the target cells. This unique capability enables NK cells to effectively eliminate virus-infected and tumor cells without prior antigen stimulation, highlighting NKp46 significance in immune surveillance and response.
