Nitrotyrosine Antibody (39B6) is a mouse monoclonal IgG2a kappa light chain antibody that detects Nitrotyrosine in non-species reactive applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-Nitrotyrosine antibody (39B6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nitrotyrosine serves as a critical biomarker for inflammation and nitric oxide (NO) production, forming when tyrosine residues are nitrosylated in the presence of reactive nitrogen species. Nitrotyrosine presence indicates oxidative stress and can be a stable product of various pathological processes, including peroxynitrite formation. Elevated Nitrotyrosine levels have been associated with several inflammatory conditions, such as septic shock, rheumatoid arthritis, celiac disease, atherosclerotic plaques, and chronic renal failure, making Nitrotyrosine an important target for research into these diseases. Understanding Nitrotyrosine′s role in these contexts can provide insights into NO-dependent damage mechanisms and overall inflammatory response, highlighting Nitrotyrosine′s relevance in both clinical and research settings.

Thank you for your question. This antibody against Nitrotyrosine is not species dependent. It will detect nitrosylated tyrosine containing proteins of any species.

Is it necessary to use a mouse on mouse kit when staining mouse tissues? I've tried this antibody on 5um mouse brain tissue sections multiple times and have only ever gotten background (IF). Can any suggestions be made?

Thank you for your question. It may not always be necessary to use a mouse on mouse kit for staining mouse tissues. It depends on your secondary antibody, and whether or not you are staining mouse tissues that express high levels of endogneous IgG. This Nitrotyrosine antibody has extensive publications for IHC/IF in mouse tissues that can be referenced. We also offer this antibody directly conjugated to different fluorochromes. Using a directly conjugated primary antibody will eliminate the need for using any anti-mouse IgG secondary antibody, and possible background or non-specific staining from the secondary antibody alone.

Hola, Este anticuerpo no es específico para una especie así que trabajará para la rata. Desafortunadamente, ya no ofrecemos los kits de tinción ABC. I need to use this Nitrotyrosine Antibody (39B6): sc-32757 for detection in samples of rat origin, will it work?

Thank you for your question. Yes, it will work for rat. This antibody is recommended for detection of nitrosylated tyrosine containing proteins, regardless of species.
