NIPA Antibody (A-12) is a mouse monoclonal IgG2a antibody that detects NIPA in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). NIPA, or Nuclear Interaction Partner of ALK, plays a pivotal role in the regulation of cyclin B1, a key protein that drives the entry into mitosis. Located primarily in the cytoplasm during interphase, NIPA is phosphorylated during the G2 and M phases of the cell cycle, which facilitates the accumulation of cyclin B1 in the nucleus just before mitosis. The phosphorylation state of NIPA is crucial, as it determines its ability to bind to the SCF complex, an E3 ubiquitin ligase that targets cyclin B1 for degradation. When NIPA is dephosphorylated, NIPA forms the SCFNIPA complex, leading to the ubiquitination and subsequent proteasomal degradation of cyclin B1. An absence of NIPA results in the abnormal accumulation of cyclin B1, which can trigger premature mitotic entry, loss of checkpoint control, and genomic instability—conditions that are often associated with cancer. Furthermore, phosphorylated NIPA may also play a role in apoptotic signaling pathways, highlighting NIPA′s importance in maintaining cellular homeostasis and preventing tumorigenesis. Anti-NIPA antibody (A-12) is an essential tool for researchers studying cell cycle regulation and its implications in cancer biology.
