SEE ALSO...

Nibrin Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects Nibrin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Nibrin antibody (B-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nibrin plays a crucial role in DNA damage response, particularly in repairing double-strand breaks, which are among the most lethal forms of DNA damage. Nibrin functions as part of a complex with Mre11 and Rad50, forming the MRE11/RAD50/NBS1 (MRN) complex that is essential for recognizing and processing DNA breaks. Nibrin′s importance is underscored by its absence in Nijmegen breakage syndrome (NBS) patients, who exhibit increased sensitivity to ionizing radiation and predisposition to cancer due to defective DNA repair mechanisms. Nibrin′s interaction with other proteins, such as Rad51 and Rad52, further highlights its integral role in maintaining genomic stability. Rad51 is vital for homologous recombination, while Rad52 facilitates the loading of Rad51 onto DNA, making Nibrin a key player in orchestrating DNA repair processes.

What application is the blocking peptide sc-515069 P appropriate for?

Asked by: cjMara
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 5 out of 5 by from GoodGood antibody for western blot (1:1000 dilution) in lung cancer cell lines
Date published: 2018-07-19
Rated 5 out of 5 by from Good antibodyWorked in rat cell line. Used in WB at 1:1000. Saw some nonspecific bands but overall worked well.

Rated 5 out of 5 by from Nibrin (B-5)Excelent antibody for western blot. Detected Nibrin at expected size in 15 ug total proteine lysate, wet transfer one hour at 450 mAmps on PVDF membrane, overnight blocking with 5% nonfat dry milk in PBS, one hour incubation at RT with 1:500 anti Nibrin in 5% PBS with 0,2%Tween followed by one hour incubation with 1:10.000 anti mouse (sc-358914) HRP conjugated antibody and detection with ECL prime substrate followed by short film exposure (under 1 minute) and automatic processing.

Rated 5 out of 5 by from Good oneI used the sample antibody to detect NBS1 by using WB in HEK-293T cells treated with doxorubicin. As expected, doxorubicin promoted NBS1 expression and the antibody did a great job showing a sharp band.

Rated 5 out of 5 by from Works very wellWorked great for us. Strong signal and only one band even with long exposure. Picture is of U2OS cell lysate (15ug) on PVDF. I used 1:2000 dilution of primary antibody (less should work fine).
Date published: 2017-08-09
Rated 4 out of 5 by from GOOD ANTIBODYThe antibody signal is strong and clear. Though few faint non specific bands can be seen
