NHERF-1 Antibody (A-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects NHERF-1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-NHERF-1 antibody (A-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NHERF-1, also known as EBP50, plays a crucial role in regulating the Na+/H+ exchange protein (NHE3), which is essential for transepithelial sodium absorption, primarily in the intestinal and renal brush border membranes. By interacting with NHE3 through its two PDZ domains, NHERF-1 facilitates the cAMP-mediated inhibition of NHE3, thereby decreasing sodium absorption, which is vital for maintaining electrolyte balance and fluid homeostasis in the body. Additionally, NHERF-1 acts as a scaffold for a multiprotein complex that includes Ezrin and NHE3, promoting cAMP-mediated phosphorylation and subsequent inhibition of NHE3 activity. The amino-terminal PDZ domain of NHERF-1 is critical for dimerization in vivo, while phosphorylation at Ser 289 by G protein-coupled receptor kinase 6A enhances regulatory functions. Furthermore, NHERF-2, also known as E3KARP, is a related protein that is ubiquitously expressed and plays a role in the regulation of NHE2, highlighting the interconnectedness of these regulatory pathways in cellular ion transport.
